چکیده
Fig. 18 Shown are the regulatory elements characterized so far, which control the transcription of the tyrosine aminotransferase (TAT) gene. The TAT gene may be induced either by glycocorticoids or by cyclic adenosine monophosphate (cAMP), which activates the protein kinase A. All distances are given in kilobases (kb) and refer to the starting point of the TAT gene transcription (arrow). mediated by cell-cell contacts play an important role. Furthermore, hormonal signals coordinate differentiation processes at the level of the organism. A close relation between signal transduction and cell-type-specific transcriptional control is exemplified by the tyrosine aminotransferase (TAT) gene which is expressed exclusively in liver cells and which is switched on briefly after birth. This switch is part of a terminal differentiation process by which the liver acquires gluconeogenic capacity as a response to specific metabolic requirements of the newborn organism.

The cyclic adenosine 3',5'-monophosphate- and the glucocorticoid-dependent enhancers are targets for insulin repression of tyrosine aminotransferase gene transcription.
The pathway of gluconeogenesis is activated in liver shortly after birth and is controlled by glucagon and glucocorticoids, which stimulate, and insulin, which inhibits, the expression of genes coding for gluconeogenic enzymes. To understand the molecular basis of this cell type-specific and coordinate control, we analyzed the cis-regulatory elements of the tyrosine aminotransferase gene, which...

The phosphoenolpyruvate carboxykinase (PEPCK) gene is regulated at the transcriptional level by a variety of effectors in a tissue-specific fashion. In order to study the parameters involved in the tissue-specific hormonal regulation of the PEPCK gene, we have used a transient expression test in well-differentiated rat hepatoma cells as well as in dedifferentiated variants. In this test, the PE...

A single far-upstream enhancer is sufficient to confer hepatocyte-specific, glucocorticoid- and cyclic AMP-inducible periportal expression to the carbamoylphosphate synthetase I (CPS) gene. To identify the mechanism of hormone-dependent activation, the composition and function of the enhancer have been analyzed. DNase I protection and gel mobility shift assays revealed the presence of a cyclic ...

Methionine adenosyltransferase (MAT) is an essential cellular enzyme which catalyses the formation of S-adenosylmethionine, the principal methyl donor and precursor for polyamines. In mammals, two different genes, MAT1A and MAT2A, encode for liver-specific and non-liver-specific MAT respectively. We previously described a switch in the MAT expression from MAT1A to MAT2A in human liver cancer, w...
متن کاملLiver enriched transcription factors and diVerentiation of hepatocellular carcinoma
The development of a complex organism relies on the precise temporal and spacial expression of its genome in many different cell types. The unique phenotype of hepatocytes arises from the expression of genes in a liver specific fashion, which is controlled primarily at the level of mRNA synthesis. By analysing DNA sequences implicated in liver specific transcription, it has been possible to ide...
